Several parameters that should be paid attention to when purchasing a lyophilizer machine
1. Freeze-dried area
Users should determine how much freeze-drying area to use according to their own needs. Hawach’s ordinary -80°C mini freeze dryer machine, FD-80 -A has drying area of 0.12㎡. For example, for each batch of products that require lyophilization of 1.2 kg (liter) of liquid, use the material tray to load the material, each tray is loaded with 10㎜ thick, then the load area of the freeze-dried board layer can be calculated:
A (Area, ㎡) = V (Volume, m3) / H (Height, m) = 0.0012m3 / 0.01m = 0.12㎡
It is necessary to select a lyophilizer machine with a layer load area of 0.12㎡.
2. Cold trap temperature
Cold trap is a device to capture water in the freeze-drying process. Theoretically, the lower the temperature of the cold trap, the stronger the water-capturing ability. The temperature of the cold trap of the laboratory lyophilizer mainly has several grades such as about -45 ℃, about -60 ℃, and about -80 ℃. Freeze-drying with the first grade temperature is suitable for some products that are easy to freeze-dry; the second for most products, and the third for special products.
3. Cooling rate
The cooling rate reflects the cooling capacity of the refrigeration system. Under no-load conditions, the cold trap temperature should reach the minimum temperature specified in the index within 1 hour. For example, for a freeze dryer with a cold trap temperature ≤-80 ° C,
(Hawach’s low cost freeze dryer -80 series), the machine will start timing when the refrigeration is turned on, and the time for the cold trap temperature to reach -60 ° C shall not exceed 1 hour.
4. Temperature uniformity and flatness of the material shelf
Temperature uniformity and flatness of the shelf have a great influence on product quality uniformity. The temperature control of the shelf includes heater type and intermediate fluid type. And the latter is better regarding the two indicators. The cooling and heating are achieved through the circulation of the intermediate fluid in the fluid channel inside the shelf, so the temperature of the layer is uniform. The temperature control of the shelf of the bell-type lyophilizer machine (Hawach’s pilot scale lyophilizer -80 series, for example) is basically a heater, and the temperature consistency of the shelf is slightly worse. But in general, the temperature difference of the shelf for medicine should be controlled within ± 1.5 ℃, and the temperature difference within the plate should be ± l ℃.
5. Control system
The control system and functions of freeze dryers are different. For the laboratory lyophilizer, it is mainly used in the exploration of lyophilization process and a small amount of trial production. Therefore, the control system should be able to display and automatically record the parameters of the freeze-drying process in real time; able to set, modify and effectively execute the freeze-drying process program; have a USB interface for data collection and storage.
